Resarch and development for maintaining and improving brain and mental health

  • The Smart Aging International Research Center (SAIRC) was founded on 1 October 2009. This Center proposes the creation of a new field to produce well-rounded personnel who have gained “totally comprehensive intelligence” from their experience, in order to cope with various problems associated with the diverse and complex super-aging society. To this end, the Center will promote interdisciplinary cooperative research, international collaborative research projects, and research collaborations with industry, as well as disseminate and exchange information by holding international conferences and symposia.  The SAIRC is designed to develop an approach for maintaining and improving brain and mental health in a diverse and complex society from the standpoints of brain science, with the aim of maintaining and improving brain functions; cognitive neuroscience and psychology, with the aim of maintaining a healthy and tranquil state of mind at each aging stage; and philosophy, phenomenology and ethics that provide a radical reconsideration of the concept of the “mind.” We welcome collaborative researches with companies, and academic consultations.

Image Guided Local Drug Delivery for Central Nervous System Disorders

  • Effective drug delivery to central nervous system is promising as a new therapeutic strategy against various central nervous system disorders. However, it represents a challenge because systemic delivery is always hindered by blood brain barrier, and local delivery cannot be effective because of the restricted distribution. Recently, local drug delivery system that can achieve robust distribution of the infusates by maintaining a pressure gradient at the tip of the infusion catheter during infusion has developed. Although this method is promising, there still exist several problems as loss of infusates by backflow through catheter tract or inconsistent distribution. We recently developed a method to deliver agents with this method under MRI guidance, and now performing clinical trial to treat recurrent gliomas.

Focal central nervous system disorders such as tumor, epilepsy, and parkinson's disease can be the target for application of this technique. Developments of the new catheters and pre-infusional similation systems are on their way. We are searching for effective collaborative research in this field.

  • We will develop a brainmorphic computing hardware, which realizes the brain-specific functions such as conscious/sub-conscious process, self, selective attention, and so on, by directly using inherent physics and dynamics of constituent devices. The resulting hardware would be small, efficient, high-performance. Some examples include the chaotic neural network reservoir, optimization through high-dimensional complex dynamics, and neural network composed of spin-orbit torque nano-devices.
  • The resulting hardware is suitable for the edge AI which learns users’ personal behavior. Examples include watching service devices embedded in hearing aids or dental implants, which monitor and learn personal cardiac and brain-wave signals or saliva ingredients, to detect abnormal situations.

Hormone Actions in Human Breast Carcinoma

  • Breast cancer is one of the most common malignancies in women worldwide. Therefore, it is very important to investigate biological features of breast carcinoma in order to improve clinical outcome of the patients. It is well known that estrogens play important roles in the development of human breast carcinomas, and endocrine therapies are frequently used in these patients to block the intratumoral estrogen actions. In the Division of Pathology and Histotechnology, we analyze hormone actions in breast carcinoma by pathological methods as well as various molecular biological techniques.

Molecular studies on new resources for hybrid rice breeding

  • F1 hybrid breeding is one of the most advanced techniques for production of cultivars with higher yield and desirable traits of both parents. Most of hybrid rice utilize cytoplasmic male sterility (CMS) and fertility restoration system. We develop a Tohoku University-original CW-type CMS and fertility restorer line and study their molecular mechanism. The CW-type cytoplasm enables indica rice cultivars to be CMS, which could not be achieved by other known cytoplasm.

Hybrid rice has an average 30% yield advantage over inbred lines, and is cultivated on 13% of the world rice fields. Our study provides a new and original resource for hybrid rice breeding for globalization of rice industry.

  • Our research group has succeeded in synthesizing various metal oxide nanoparticles with controlled size and exposure crystal planes by using organic modifiers under supercritical water conditions. The oxygen storage/release capacity of those materials in the low-temperature region is very high, and the reforming reaction of oxidative hydrocarbon proceeds at a significant rate. Besides, by combining the supercritical CO2 drying method, we have succeeded in forming a complex in which oxide nanoparticles are dispersed at a high concentration on the surface of the porous material, realizing both high oxygen storage/release capability and stability.

Low-temperature reforming reaction of biomass wastes, heavy oils, and methane. In the future, it is expected to be a technology that will lead to the construction of a low-carbon society, including CO2-free complete recycling of waste plastics.

Development of Recombinant Inbred Mice with a Genetic Predisposition to Collagen Disease

  • Eleven strains of recombinant inbred (RI) mice derived from MRL/lpr and C3H/lpr mice were established. This RI is the only one in the world that randomly develops lesions such as nephritis, arthritis, sialadenitis, vasculitis, and production of autoantibodies in each strain. The genomes of the two strains of mice are randomly held in homozygous condition, and the phenotypes of each strain and the effects of administered drugs could be compared based on their genotype maps. It is possible to identify the regions of gene loci involved in the phenotype and drug sensitivity.

High-Precision Capturing of 3-D Dental-Arch Shapes and Their Occlusal Relation and its Potential Implementation into CAD/CAM Dentistry

  • In spite of the long history of digital dentistry, the accuracy of the prosthetics fabricated with CAD/CAM systems is far from satisfactory. Jaw opening during taking impression elicits deformation of the mandible and the lower dental arch, and consequently causes the inaccuracy of occlusal relationship of the upper and lower dental arches on the CAD data. The method presented here utilizes the check-bite record at the maximum intercuspation to correct the relative positions among the teeth in the dental arches as well as between the arches. After this correction, the accuracy of the CAD data is satisfactory enough for the fabrication of the dental prosthetics which need no adjustment during their setting.

This method uses the newly-developed check-bite recording technique together with the conventional precious impression or optical impression techniques. It can easily be applied to the commercial CAD/CAM systems in the market. We hope to collaborate to the manufacturers of dental CAD/CAM systems to increase the accuracy of the systems.

  • Angiogenesis, i.e., the formation of new capillaries, takes place under pathophysiological conditions including cancers, ocular neovascular diseases and atherosclerosis. Anti-angiogenic drugs have been developed, and are now in clinical use. However, because of side effects due to the damage to vascular endothelial cells, the development of anti-angiogenic drugs with different modalities is anticipated. We have isolated vasohibin-1 (VASH1) as a novel angiogenesis inhibitor expressed by endothelial cells (ECs), and vasohibin-2 (VASH2) as its homologue having pro-angiogenic activity.

We are planning to develop strategies of VASH1 potentiation and VASH2 inhibition. For VASH1 potentiation, we are planning to use recombinant VASH1 protein or search low molecular weight substances having VASH1 like activity. For VASH2 inhibition, we are succeeded in the development of neutralizing anti-human VASH2 monoclonal antibody.

Development of catalytic reaction system for direct synthesis from CO2 and diols

  • We found an effective catalyst system composed of CeO2 and 2-cyanopyridine for direct and catalytic polycarbonate formation reaction from CO2 and diols. CeO2 acted as an activator for CO2 and diols, and 2-cyanopyridine shifted the equilibrium in favor of the product side by hydration of 2-cyanopyridine, promoting the reaction. Green polycarbonates can be synthesized by combination of the catalyst system with a technique of diol syntheses from biomass.

This catalyst system is effective for direct transformation of CO2, contributing to the effective utilization and emission reduction of CO2. Combination of the catalyst system with a concentration technique of CO2 will bring about much advantage.
