Development of Fish iPS Cell Technology

In order to utilize for the development of fish iPS cells, we established experimental systems to monitor cellular multipotency using zebrafish and medaka transgenic lines which express GFP fluorescence under the regulation of oct4 promoter. Because fish has strong fin regenerative activity after amputation, and GFP positive cells appear during fin regeneration, fin is expected to be suitable for iPS cell preparation. In addition, we ascertained that MC-LGNSO, a commercially available minicircle DNA developed for human iPS cell preparation, works well in fish cells. By using transgenic lines and the minicircle DNA, we are now trying to establish iPS cells from regenerating fin cells.Targeted Application(s)/Industry
We expect that in fish, iPS cell technology is applicable to innovative new technologies in aquaculture: i.e. generation of iPS cells from selected fish strains and production of clone individuals of the selected lines, and cryopreservation of important fish strains as cell lines.Researchers
